Objectives: 2-methyl-4, 5-diphenyl-1H-immidazole had been successfully synthesized. This was proven by the analysis results of 1H and13C NMR, and the analysis of functional groups using FTIR. The ligand obtained was complex with metal ion cobalt (II), ion Mn (II) and ion Cu (II). Methods/Analysis: The complex formula produced was based on the characterization using CHN element contents analysis, FTIR and thermogravimetry analysis. When 2-methyl-4, 5-diphenyl-1H-immidazole ligand was complex with cobalt (II) ion, complex formula [CoL2Cl2 (H2O) 2] ∙3,5H2O (L=2-methyl-4, 5-diphenyl-1H-immidazole) was showed. The LC50 obtained using Brine Shrimp Lethality Test on cobalt (II) complex was 168, 24 ppm. When 2-methyl-4,5-diphenyl-1H-immidazole ligand was complex with manganese(II) metal ion, a complex in the shape of thebrown-black polycrystalline complex was formed with a yield of 73,24%. Findings: The complex formula produced based on the characterization using AAS, CHN analyzer, FTIR and TGA was MnL (H2O) 3Cl2·H2O (L=2-methyl-4, 5-diphenyl-1H-immidazole). Synthesized manganese (II) complex toxicity test using Brine Shrimp Lethality Test produced an LC50 value of 38, 03 ppm. When 2-methyl-4, 5-diphenyl-1H-immidazole ligand was complex with copper (II) metal ion, a red crystal solids were produced with a yield of 93, 72%. Novelty/Improvement: The characterized results of copper (II) complex using UV-Vis spectrophotometer, infrared spectrophotometer, CHN Analyzer, SSA, and TGA showed a complex formula of [CuL2Cl2]∙5H2O (L= 2-methyl-4,5-diphenyl-1H-immidazole). The LC50 value of 2-methyl-4, 5-diphenyl-1H-immidazole ligand obtained from Brine Shrimp Lethality Test was 192, 31μg/mL.
