Lysozyrne-DPPC Interactions in the Model Membrane, DPPC-H2O

The effect of the protein, lysozyme, on the hydrocarbon chain melting phase trasition of the model membrane system, DPPC-H2O/D2O, as been investigated by DSC, 1H NMR and fluorescence spectroscopy. It is found that the protein binds to the lipid headgroups and that the effective lipid headgroup-headgroup interaction is increased due to lipid-protein interaction. The tryptophan residue(s) of the proteinis implicated in thisassociation. The interaction isstronger when the membrane is in the form of vesicles rather than that of lamellar stacks. The lipid headgroup-lysozyrne binding results in reduced mobility of the acyl chains as well as the headgroup of the lipid molecule.
