Buccoadhesive bilayer tablets of Tapentadol Hydrochloride were prepared and evaluated in this investigation. Tapentadol is a centrally-acting opioid analgesic, having potency between morphine and tramadol. The direct compression method was used for preparing tablets. The polymers were employed in the formulations are Carbopol 934 P, Eudragit RL 100, sodium alginate, Chitosan, and polyvinylpyrrolidone-K30 (PVP) and ethyl cellulose (EC) as a backing layer. The prepared formulations were evaluated for their physicochemical characteristics, swelling index, surface pH, ex vivo buccoadhesive strength, in vitro, in vivo drug release and ex vivo permeation studies. The significant differences in the results were observed, which dependent on characteristics and composition of bioadhesive materials used. Stability studies were conducted in natural human saliva and accelerated conditions, results reveals no significant change in physical appearance. Drug content, buccoadhesive strength and the P-value shown statistically significant at <0.05. Ex vivo mucous irritation by histological examination reveals, the administration site of buccal tablet over the buccal mucosa did not cause any irritation, ulceration, inflammation and redness, and it resembles to controlled buccal mucosa. Good correlations were observed between in vitro and in vivo drug release, with a correlation coefficient of 0.997. Drug diffusion from buccal tablets showed apparently zero order kinetics and release mechanism was diffusion controlled after considerable swelling.Keywords

Colon specific drug delivery has gained a much importance for the delivery of drugs to treat the both local and systemic diseases. Local diseases include Chron's disease, ulcerative colitis, and colorectal cancer. Number of other serious disorders like nocturnal asthma, arthritis and angina can also be cured by these techniques. Colonic delivery is a good candidature for delivery of proteins peptides and vaccines where the enzymatic degradation and the hydrolysis of proteins can be minimized and increases the systemic bioavailability. A drug should be protected from the absorption and the upper GI environment to achieve the successful colonic drug delivery. The colon specific delivery of drugs to the target receptor sites has the advantage to reduce the side effects and improves the therapeutic response. Colon specific drug delivery are being developed by taking advantage of the luminal PH conditions and the presence of microbial enzymes such as azoreductase, pectinase, dextranase…etc. This review mainly reveals on the various concepts and approaches include Prodrug, PH and time dependent systems and microbially triggered systems used in the development of colon specific drug delivery. This also focuses on the novel approaches namely Pressure controlled colonic delivery, osmotic controlled drug delivery and CODESTM. In vitro and in vivo evaluation parameters has been discussed here.Keywords

Hyperlipidemia and high levels of reactive oxygen species in the body are key factors for the development of cardiovascular diseases such as atherosclerosis. The present study was designed to investigate the Hypolipidemic and antioxidant activity of an herbal concoction prepared from leaves of Artocarpus heterophyllus, Mimusops elengi and Talinum portulacifolium. Hyperlipidemia is induced in rats by Triton WR-1339. The concoction showed significant reduction in all serum lipid parameters, together the results showed increase in antioxidant levels and decrease in proxidant levels. Fortunately when histopathology of carotid artery is observed indicating that the concoction can also exert antithrombotic activity to some extent.Keywords
