Abstract

Aim: The aim is to develop simple validated analytical method for analysis of Tofacitinib by UV Spectroscopy and to study the forced degradation and stress conditions have been used to detect the stability of Tofacitinib. Method: Tofacitinib was estimated at 285.9nm. Linearity range was found to be 10-50 mcg/ml. The correlation coefficient was found to be 0.9996. The molar absorptivity was found to be 12468.77mol/cm. The proposed method Sandell's sensitivity was found to be 0.040410 μg cm²/0.001AU. The limit of detection and quantification were found to be 0.8169 and 2.4755 μg/ ml respectively. The degradation behavior of Tofacitinib was carried out as per the standard procedures and guidelines. Forced acid hydrolytic degradation, alkali degradation and oxidative degradation of was performed in bulk Tofacitinib and laboratory prepared admixtures using 1M Hydrochloric acid up to 48 hrs, in 10 % Hydrogen peroxide up to 48 hrs and for 1.0 M Sodium hydroxide up to 10 min at room temperature. The resulting solutions were analyzed for content by UV spectrophotometry at the maximum absorption of 285.9 n. The assay value of Tofacitinib in bulk and physical admixture was calculated at different time intervals for intraday and interday experiments. Results and Conclusions: The proposed method was successfully applied for the determination of tofacitinib in pure and laboratory prepared physical mixtures. The % RSD value of Tofacitinib in bulk and physical admixture was calculated at different time intervals for recovery , precision (Iintraday and Interday experiments) and quantification studies were found to be less than 2 %.

Abstract

OBJECTIVE: A study was conducted to evaluate drug utilization of gastrointestinal agents in pediatric and geriatric patients. METHODS: Randomly selected 150 patients of gastrointestinal diseases with 92 male and 58 female of age from 15-58 underwent drug utilization evaluation of gastrointestinal agents. The frequencies of gastrointestinal agents were calculated and compared WHO indicators of DUS (drug utilization study) guidelines. RESULTS: Results include patients on long term gastrointestinal drug therapy on geriartric patient, paediatric patient. It include 92 male and 58 female patients in which 38.66% of female patient was and 61.335 of male patient suffered from gastrointestinal diseases. Out of 150 patients21.33% of patients was treated with pantoprazole,12% of patients was treated with esomeprazole ,28.66% of patients was treated with omeprazole,10.66% of patients was treated with rabeprazole,27.33% of patients was treated with ranitidine. Oral route of administration was more than parenteral administration. Gastritis is occurred more than inflammatory bowel disease, peptic ulcer disease and gastroesophageal reflux disease

Abstract

The aim of the study is to monitor and observe the incidence of intravenous drug administration errors in the general medicine department of a tertiary care hospital. Preparation and administration of the intravenous drugs to in-patients of the general medicine department was monitored by direct observational method. In a total of 329 intravenous administrations, 54.71% of errors were observed in the patient population out of which 74.78% showed at least one error. The results revealed that the flow rate error 45% were more prevalent among the overall study population followed by reconstitution error 26%, wrong mix errors were found to be 23% and incompatibility were found to be 10%. Errors were observed in both intravenous infusions as well as in intravenous bolus injections. Since intravenous administration of drugs have advantages of quicker delivery of drug to the target site via blood, proper care should be taken by nurses and other health care professionals in the IV drug administration. Errors can be minimized by utilising advanced techniques like small infusion smart pumps and automated flow rate infusion set.

Abstract

Steganography is the art of hiding the fact that communication is taking place, by hiding information in other information. Many different carrier file formats can be used, but digital images are the most popular because of their frequency on the Internet. For hiding secret information in images, there exists a large variety of steganographic techniques some are more complex than others and all of them have respective strong and weak points. Different applications have different requirements of the steganography technique used. In this paper presents hiding the data behind the image then encrypt the image by using hyper-chaos encryption algorithm. In this algorithm, shuffling matrix and diffusing matrix are generated based on Chen’s hyper-chaotic system. Firstly, the Chen’s hyper-chaotic system is used to shuffle the position of the image pixels, and then use Chen’s hyper-chaotic system to confuse the relationship between the original image and the cipher image.

Abstract

The social networks has made it easier than ever for users to share their text file, document and other media content with anybody from anywhere. It’s easy to access the user-generated media content which brings about the privacy concerns. Traditional access control mechanisms were implemented for a single access policy is made for a specific piece of the content, cannot satisfy the user privacy requirements in large-scale media data sharing systems. Instead, configuring many levels of access privileges for the shared media data content is needed. On one hand, it determines the principle of social networks in information circulation. On the other hand, it accords with the diverse social relationship among social network users. In this paper, we propose the Scalable Media Access Control (SMAC) system to enable such a configuration in a secure and in an efficient manner. The proposed system SMAC system is permitted by the Scalable Cipher Text Policy Attribute-Based Encryption (SCP-ABE) algorithm as well as a comprehensive key management schema. Also we provide formal security proof to prove the security of the proposed SMAC system. Additionally, we conduct the intensive experiments on mobile devices to demonstrate its efficiency.

Abstract

Cataract is the major cause of blindness all over the world, nearly 42% of visual impairment occurs due to diabetic associated complications in the eye. Borreria hispida, belonging to Rubiaceae family was used to treat eye infections by folklore system of medicine in India. This study was aimed to create scientific evidence for the anti-cataract activity of hydroalcoholic leaf extract of Borreria hispida (HAEBH) in diabetes induced cataract using goat lens. Cataract was induced using galactosamine in goat eyes lens. In this study, isolated goat lenses were suspended in bovine serum albumin. Totally sixteen lenses were selected, each group containing four lenses - normal control (distilled water treated lens), galactosamine treated lens, 1mg/ml of the hydroalcoholic extract of Borreria hispida leaves extract treated lens (HAEBH) and finally 1mg/ml of standard drug atropine treated lens. The opacification of lens were monitored in all the sixteen lenses. Among these, the anticataract activity of Borreria hispida was strengthened by the reduction in the deposition of proteins which is involved in the opacification of lens compared to galactosamine treated lens. The extract shown effective anticataract effect similar to that of standard group. The hydroalcoholic extract of Borreria hispida possess anticataract activity may be due to inhibition of Aldose Reductase enzyme involved in the polyol pathway which has a prime role in developing diabetes induced cataract. Further studies has to be carried out to isolate the prominent biomolecule responsible to control diabetes associated cataract.

Abstract

Inflammatory bowel diseases (IBD) termed as chronic inflammatory disorders of the gastrointestinal tract distinct by episodes of reversion and decline. The two identified subtypes of the disease, ulcerative colitis and Crohn's Disease which differ in forms of clinical presentation. Environmental factors, including infections, diet, lifestyle factors, medication use have contributed to alterations in the global prevalence of the disease. Although the exact pathogenesis of IBD remains unknown, part of the underlying mechanism is a deregulated host immune response to intestinal flora, in genetically vulnerable Beings. The incidence of IBD is persistently rising in other earlier low incidence areas, such as Asia and the developing world. The annual frequency of CD is highest in North America (20.2 per 100,000, per person-years); whereas the annual occurrence of UC is highest in Europe (24.3 per 100,000 per person-years) Environmental risk factors of IBD is smoking, air water pollution, vitamin D, dietary, NSAID, stress. The newly described Th17 cells are also involved in the gut inflammatory answer in IBD. To understand the pathogenesis of IBD alternative model has been used such as zebrafish, c. elegans, Drosophila. The novel treatment of IBD such as vedolizumab, Etrolizumab, agents targeting specific pathways in IBD, but unfortunately possess life-threatening adverse effect and globally increased cost. This review also focuses on the favourable results from the use of herbal products in the treatment of IBD.

Abstract

Tinospora cordifolia is an herbaceous vine that belongs to the Menispermaceae family, a succulent woody shrub genus, native to the tropical Indian sub-continent, and it is known for its common names: “Guduchi”, “giloy”, “amrita”, “gurjo”, and “heart-leaved moonseed”. The recent recognition and biological role of active components in disease management in the plant have contributed to an active interest in the plant worldwide. Extracts of T. cordifolia are widely used as anti-spasmodic, anti-microbial, antiperiodic, immunomodulatory, anti-inflammatory, anti-arthritic, anti-allergic, endocrine disorders like anti-diabetic, hypolipidaemic, etc. in various herbal formulations to treat different diseases. It helps reduce the resistance to insulin and stress, revitalises all the tissues of the body, and spontaneously promotes metabolism. However, this adaptogenic ability is also under-explored. Stress and ageing disorders need to be treated independently. The current review paper mainly focused on stress, depression, and the anti-stress properties (adaptogen) of T. cordifolia. Depression and stress, along with disrupted metabolic and reproductive properties, are high-risk factors in polycystic ovarian syndrome patients. Further studies are needed to focus on adaptogenic activity to relieve certain endocrine disorders that occur due to stress, often ovarian cysts, female menstrual irregularities, and hormonal imbalance.

Abstract

The kidney plays an essential role in removing waste products and drugs from the body and maintaining balanced body fluids. It gets affected due to many factors, notably, diabetes and high blood pressure. Nowadays, naturally, derived products are essential in curing various ailments and are safe and cost-effective. The purpose of this study is to assess the toxicity profile and nephroprotective effect of a proprietary polyherbal formulation in Wistar albino rats for gentamicin-induced nephrotoxicity. The Polyherbal formulation was procured from Rumi Herbals Private Limited. Acute toxicity experiments were conducted in Wistar rats using the Gentamicin induced nephrotoxicity model as per OECD standards 423, and the efficacy was assessed using the Gentamicin induced nephrotoxicity model. The formulation was proven safe up to 2000mg/ kg orally in an acute toxicity study, with no behavioral abnormalities and no fatality. The gentamicin 80 mg/kg i.p for 7 days induced nephrotoxicity in rats showed a significant (P<0.05) increase in the renal parameters and reduction in antioxidant levels compared with day 0. Whereas test drug-treated groups at a low dose (200 mg/kg) and high dose (400 mg/kg) showed significant (P<0.05) reduction in elevated renal parameters and improvement in antioxidant levels compared with the disease control group. According to the histopathological interpretation of isolated kidneys, this formulation protects from kidney damage and restores typical kidney architecture. From the results, the proprietary polyherbal formulation has shown effective nephroprotective activity may be due to the presence of secondary metabolites/phytoconstituents. Further investigation is essential to focus on the mechanism involved and standardize the active phytoconstituents responsible for the nephroprotective activity.

Abstract

The pharmacological healing for inflammatory bowel diseases continues to be uncertain and requires immediate therapeutic interventions. A poly-herbal formulation obtained from a traditional and authentic classic text of Ayurveda was assessed for its effect against IBD (inflammatory bowel disease) in this study. The formulated poly-herbal suspension comprises three different drugs namely, Burma dhaniya (Eryngium foetidum), Sapota (Manilkara zapota), and Curry leaves (Murraya koenigii). The formulated suspension was evaluated for certain standard parameters like organoleptic and accelerated stability studies at various temperatures. It was checked for its efficacy by oral route in acetic acid-induced colitis affected Balb/c mice. Mice were orally administered with formulated suspension (275 mg/kg, 550 mg/kg,), every 24 hours for 10 days. Histopathology, macroscopic damage score, myeloperoxidase (MPO) activity, and red blood cell parameters were evaluated after treatment. Reduction in the MPO activity, decrease in the macroscopic damage scores, and an increase in RBC cell count were seen distinctly at a high dose of 550 mg/Kg. The results obtained, established the effectiveness of the poly-herbal suspension against inflammatory bowel disease by treating the mice from acetic acid-induced colitis by reducing inflammation and oxidative damage to the colon. The maximum therapeutic effective activity was found to be 550 mg/kg for IBD mice.

Abstract

The valued studies of alternative psoriasis treatment options are in a much higher need among the Scientific Community. This study aimed to evaluate the anti-psoriatic activity of ursolic acid cream in imiquimod-induced psoriasis in BALB/c mice. The creams containing ursolic acid, a pentacyclic triterpenoid at percentages of 0.1 and 0.2% were formulated. The pH, spreadability, physical characteristics and acute dermal irritation of the cream were assessed. Animals were grouped into five each having 6 animals. Clobetasol, a topical corticosteroid, was used as the standard. One group was used as control and four groups were treated with the formulated imiquimod cream while receiving treatment. Parameters such as skin inflammation severity, ear thickness, plasma level of interleukins (IL)-17, histology of the back of the skin and spleen weight were evaluated. Erythema and scales were scored on a daily basis with the 0.1 and 0.2% ursolic acid cream significantly ameliorating psoriatic-like symptoms in a manner comparable to clobetasol. Imiquimod-induced epidermal hyperplasia and inflammation were inhibited by topical application of ursolic acid as shown by the results of histopathology. Spleens of the positive control group were larger in comparison with the rest of the groups. BALB/c mice treated with ursolic acid creams exhibited a decrease in the plasma levels of cytokines IL-17 when compared to the positive control group. The result of this study provided an insight that topical application of ursolic acid can be a potential treatment for psoriasis.
