- Correlation between Osteopontin Promoter Gene and Fresh Semen Quality in Friesian Holstein Dairy Cows

This study aims to investigate the polymorphism of osteopontin promoter gene in Holstein Friesian (FH) dairy cows and its correlation with quality of fresh semen in FH dairy cows. The basis determining for osteopontin as the main bio-marker in the determination of male Holstein dairy cow fertility by several previous studies which showed that osteopontin levels in seminal plasma Holstein dairy cows with a good fertility have osteopontin concentrations higher than the low fertility of dairy cows. A total of 14 blood samples taken from Holstein Friesian dairy cows then the DNA extracted and amplified using primers SPP1F and SPP1R. The 306 bp band as a target was detected in all 14 samples then sequenced for analysis of the nucleotide base. The results showed that all samples with low sperm concentration occurred deletion at10080 bp and the transition (G-A) at 10090 bp. The results indicated that this mutation site could be related with the trait susceptibility to sperm concentration. Further studies are needed to address other parameter related to comprehensive sperm quality.Keywords

Pollutants include particulate matter suspended referred to as Particulate Matter (PM). Soot is one of the components of PM that are known to be cytotoxic and causing the occurrence of inflammation in cardiovascular and respiratory systems and can pass through the placenta barier may affect to the fetus. Effect by pollutants depends on the magnitude of exposure and length of time of exposure. This research aims to identify the Caspase-9 as an indicator of apoptosis on placenta white rats pregnant (Rattus norvegicus) is being exposed carbon black with immunohistokimia techniques and cell number tropoblas due to exposure to carbon black. This research begins with a fecundate female rats and exposure with carbon black dosage 534 mg/m3 and 1064 mg/m3. The next step is the observation and examination of the expression of Caspase-9 by immunohistokimia staining of white rat placenta that had being with carbon black. The design of this research using design random group who were conceived to prove the existence of expression caspase-9 as an indicator that mediate apoptosis in white rats pregnant is being exposed carbon black. The number of cells tropoblast analyzed by using analysis of variant (ANOVA) and the difference between the control group with treatment in a test using LSD test. The result of expression Caspase-9 in pregnancy 6-17 day with exposure to carbon black doses 1064 mg/m3 shows the results of significantly. It showed that the age of pregnancy affect the expression Caspase-9. On the outcome of research on pregnancy day to 6-11 and day 6-17 show a significant difference when a compare to control both on a cell cytotrophoblast and synsitiotrophoblast. Conclusion this research is that influence exposure carbon black on the placenta rats white (Rattus norvegicus) with long exposure to 6-11 the during the pregnancy, doses 534 mg/m3 and 1064 mg/m3, increase expression protein Caspase-9 as an indicator mediate apoptosis along with long exposure to and increased doses and decrease the total cell tropoblast placenta due to exposure to carbon black along with increasing long and dosage of exposure.Keywords
