Cytotoxic Activity of Mangosteen (Garcinia mangostana L.) Peel Extract and α-Mangostin toward Leukemia Cell Lines (HL-60 and K-562)

Abstract

Fruit of Mangosteen (Garcinia mangostana L.) is well-known in Indonesia and other Southeast Asian countries. Studies have shown that extract of the pericarp of mangosteen contained mostly of xanthones exhibit many biological activities, especially as an antitumor. This study aimed to investigate the cytotoxic activity and selectivity of Mangosteen Peel Extract (MPE) and α-mangostin against two leukemia cell lines (HL-60 and K-562) and the normal lymphocyte cells from two different donors. The cytotoxic activity was performed using 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxyme-thoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay. Imatinib and Isotretinoin were used as a positive control to the K-562 and HL-60 cells, respectively. The MPE and α-mangostin revealed higher mortality toward leukemia cell lines rather than toward lymphocyte cells, with more than 80% of HL-60 and K-562 cells died at 6.25 and 25 μg/ml, respectively. MPE was more toxic and selective against K-562 with IC50 of 2.79 μg/ml and SI of 8.27, while α-mangostin was more toxic and selective against HL-60 with IC50 of 1.12 μg/ml and SI of 22.34. MPE and α-mangostin showed potent sensitivity and selectivity to leukemia cells, hence these are considered as promising sources for future leukemia treatment.

Abstract

Inflammation is one of the important biological responses to injury. Anti-inflammatory is therefore proposed to treat both acute and chronic inflammation. Chemical compounds of various plants are widely used in treatment of inflammation. Objective: This study aims to evaluate anti-inflammatory potential of G. vulgaris extract (GVE) and A. muricata extract (AME) on LPS-stimulated murine macrophage cell line (RAW264.7). Cell viability assay to evaluate nontoxic concentration in cell line was performed with MTS assay. Parameters to determine anti-inflammatory activity between treatment group and non treated cells, were IL-1β, TNF-α, and IL-6 which was measured with Elisa, and NO level which was measured with nitrate/nitrite colorimetric assay. Both GVE and AME of 50 and 10 μg/mL showed high viability (>90%) and it was not significantly different compared to control, makes it suitable for treatment. GVE and AME of 50 μg/mL resulted low TNF-α level in RAW264.7(313.16pg/mL and 264.69 pg/mL respectively), as well as IL-1β level (903.53 pg/mL and 905.00 pg/mL respectively) and IL-6 (175.88 pg/mL and 219.13 pg/mL respectively). Whereas, GVE and AME of 75 μg/mL showed lower NO level (9.76 μM and 9.79 μM respectively) compared to untreated cells. This research revealed that GVE and AME possess the anti-inflammatory potential indicated by inhibition of inflammatory mediators including TNF-α, IL-1β, IL-6 and NO.

Abstract

Aging is a natural process in humans as accumulation of oxygen-derived free radicals which leads to the activation of hyaluronidase, collagenase and elastase, that can further contribute to cellular and tissue damage. Bioactive compounds from plants have been used as antioxidant that might inhibit aging processes as well. This study aimed to determine antioxidant and anti aging properties of Oryza sativa Extract (OSE), and its compounds, vanillin and coumaric acid. The phytochemical analysis of OSE was performed with Farnsworth modified method. Antioxidant activities were performed by measurement of 2,2-diphenyl 1-pichylhydazyl (DPPH) free radical scavenger, Ferric Reducing Antioxidant Power (FRAP), and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) reducing activity, while anti aging assay were observed through inhibitory of elastase, collagenase, and hyaluronidase activities. Phytochemical analysis showed the presence of terpenoids and saponins in high level. OSE showed lowest DPPH activity (IC₅₀ = 314.51 μg/mL) compared to vanillin (IC₅₀ = 283 μg/mL) and coumaric acid (IC₅₀ = 255.69 μg/mL). In ABTS assay, OSE resulted lowest activity(IC₅₀ = 145.67 μg/mL), compared to vanillin (IC₅₀ = 4.96 μg/mL) and coumaric acid (IC₅₀ = 1.67 μg/mL). OSE also showed the lowest FRAP-reducing activity (21.26 μM Fe(II)/μg), compared to vanillin (35.05 μM Fe(II)/μg) and coumaric acid (48.52μM Fe(II)/μg). OSE showed the lowest collagenase, elastase, and hyaluronidase inhibitory activity (IC₅₀ = 816.78,107.51, and 203.13 μg/mL), compared to vanillin (IC₅₀ = 16.27, 14.46, 45.23 μg/mL respectively) and coumaric acid (IC₅₀ = 146.89, 25.38, 8.21 μg/mL respectively). In summary, OSE possess the lowest antioxidant and anti aging activities compared to vanillin and coumaric acid.

Abstract

Obesity is one of the risk factor for dyslipidemia incident, cancer, diabetes mellitus, and cardiovascular disease. Treatment of obesity using common commercial drugs shows low rate of success, hence natural products may provide better or more efficient therapeutic approach. Mangosteen (Garcinia mangostana L.) (Clusiaceae) peel extract contains xanthones and it is potentially used as alternative medicine for obesity. This research was done to determine the anti-obesity characteristics of Mangosteen Peel Extract (MPE) and its xanthones α-Mangostin (AM) and γ-Mangostin (GM) on 3T3L1 cell line. Anti-obesity effects of MPE and xanthones were investigated using differentiated-3T3L1 preadipocyte cells. Inhibitory activity of the extract and compounds on the production of Triglyceride (TG), Cholesterol (CHOL), Glucose-6-Phosphate Dehydrogenase (G6PDH) activity, and lipid droplets were examined. MPE and its compound were capable to inhibit the production of TG, CHOL, G6PDH, lipid droplets. MPE 50 μg/mL and GM 75 μM were the most active to lower TG 57.95% and 59.72%, CHOL 33.33% and 31.68%, G6PDH 52.90%, 41.95%, lipid droplets 72.99% and 70.07% respectively. In conclusion mangosteen peel extract and γ-Mangostin are the most active antiobesity compared to α-Mangostin.

Abstract

Mangosteen (Garcinia mangostana L.) is one of the most popular fruits which has been widely used medicinally. The major constituents are mostly found in the pericarp particularly, xanthones, which are tricyclic isoprenylated polyphenols. Xanthones have been reported for its anti-oxidant, anti-inflammatory, anti-bacterial, anti-proliferative, proapoptotic and anti-carcinogenic activities. In this study, the mangosteen pericarps in various maturity levels were investigated using various characterization techniques such as PSA (Particle Size Analyzer), Scanning Electron Microscope (SEM), X-ray Diffraction (XRD), and X-ray Fluorescence (XRF). The results revealed the characteristic of mangosteen at various maturity levels.
