Abstract

Three month old female mice were ovariectomised and after 4 days the mice were separated into six groups, I) ovariectomized, II) ovariectomized + D-galactose treated, III) ovariectomized + D-galactose treated + Tween 80, IV) ovariectomized + D-galactose&centrophenoxine treated, V) ovariectomized D-galactose treated + olive oil injected, and VI) ovariectomized + D-galactose treated + estrogen injected. After 15 days the mice were sacrificed. The protein concentration, amylase activity, lipid peroxidation and fluorescence product were measured in various salivary glands, viz., submandibular, sublingual, and parotid. In ovariectomized (group II) and ovariectomized + D-galactose treated (group III) mice the protein concentration and amylase activity of various salivary glands decreased, while lipid peroxidation and fluorescence product increased. In ovariectomized + D-galactose + centrophenoxine treated (group IV) and ovariectomized + D-galactose treated + estrogen injected (group VI) mice protein concentration and amylase activity in the salivary glands increased, while lipid peroxidation and fluorescence product decreased. These results reveal that centrophenoxine, and therefore aging, has modulatory effects on salivary glands in D-galactose induced aged female mice.

Abstract

The effect of testosterone on glycoprotein synthesis in salivary glands has been worked out. When salivary gland sections were stained for Periodicacid (PAS) and Alcian blue (AB) techniques, it was observed that glycoproteins were mainly located in the secretory acini of salivary glands. The staining intensity was reduced in castrated mice and increased after the injection of testosterone (4mg/250g body wt.). Soluble glycoproteins from salivary glands were isolated from defatted powder of the glands wi1;h 1 M CaClj. In juveniles, concentrations of hexoses and sialic acid were low, increased in juvenile adults and were maximum in adults. In adult castrated mice, these concentrations decreased and upon testosterone propionate administration, they increased. Soluble glycoproteins were separated on 5% polyacrylamide gel electrophoresis. With AB pH 2.5 and AB pH 1 there were 2 bands for the submandibular gland for glycoproteins and 8 and 9 bands for AB pH2.5 and pH 1.0, respectively. In both the glands some bands completely disappeared and some of them were reduced in their intensity in castrated mice and intensity was regained after the injection of testosterone. The results suggest that testosterone plays an important role in the synthesis of glycoproteins in acinar cells of salivary glands.

Abstract

One and three months old mice of both the sexes were sialoadenectomised and maintained under normal condition in the animal house upto the age of three and six months, respectively. During this period, their weight was recorded. Alloxan monohydrate was used to induce diabetes in some of them. Twenty four mice which were sialoadenectomised were subjected to fasting for 24 h and 36 h. Blood glucose, liver glycogen and alkaline phosphatase activity in liver, soleus muscle and kidney were determined in experimental and normal mice. The blood glucose level was considerably decreased in sialoadenectomised and sialoadenectomised diabetic mice compared to normal and diabetic cases. The decrease was much higher in the sialoadenectomised mice which were denied food for 24h and 36h. Glycogen content was decreased significantly in sialoadenectomised and sialoadenectomised diabetic mice. But in fasted sialoadenectomised mice, there was f6ss decrease than .fasted control. In sialoadenectomised and sialoadenectomised diabetic mice, alkaline phosphatase activity was decreased. The decrease was highly significant in liver and soleus muscle.
